研究內容概述:
The goal of my PhD thesis is to design an extraction and amplification module for viral RNA. I intend to purify RNA form HCV, do the reverse transcription to obtain the cDNA and amplify the cDNA using ccPCR. The amplification cycle entails of denaturing the cDNA at 950C, annealing the primer at 500C and finally extension at 750C. However, ccPCR employs a single temperature control at the base of a capillary tube and using free convection, the reagents circulate passing through the different temperature regimes optimal for the above named reactions.
However, we intend to use simulation to gain a foresight of the temperature distribution and the time reagents have for each of the above reactions. This will aid in our capillary tube design to optimize the use of the reagents while still maintaining the simplicity of the system.
We intend to use an in-house code specifically designed for above challenge. The code is written in C++ and parallel using OpenMP on a shared memory platform.
